Overall Project Objective: To determine the reliability of three molecular biology-based tests for Mycoplasma fermentans (incognitus strain) in human pure blood mononuclear cells. These tests include 1) "usual" polymerase chain reaction procedure (UPCR), 2) an optimized chelex polymerase chain reaction (OCPCR) procedure, and 3) a nucleoprotein gene tracking (NGT) procedure. Status/Results to Date: The analyses have been completed, and indicate that there is no greater than chance reliability between laboratories and within laboratories, for all three tests. Currently, disagreements among the investigators regarding the interpretation of results remain. An independent group of experts reviewed the protocol, data, results, and a draft manuscript and concluded that additional research should be conducted before these data could be adequately interpreted. We found that immediate family members of veterans diagnosed with Gulf War Illnesses (GWI) often complain of fatiguing illnesses, and upon analysis they report similar signs and symptoms as their veteran family members, except that their children are often diagnosed with Autism. Since a relatively common finding in GWI patients is a bacterial infection due to Mycoplasma fermentans , we examined military families (149 patients: 42 veterans, 40 spouses, 32 other relatives and 35 children with at least one family complaint of illness) selected from a group of 110 veterans with GWI who tested positive (~42%) for mycoplasmal infections. The Mycoplasmas Introduction: The first of these curious organisms was isolated in 1898 by the French scientists, Nocard and Roux from the pleural fluids of cattle with an infectious disease called bovine pleuropneumonia. After the discovery of Pleuropneumonia organisms, numerous other organisms resembling them were reported. These organisms were called pleuropneumonia-like organisms or PPLO. These organisms are now referred to as mycoplasmas, and they are the smallest and simplest free-living bacteria lacking a rigid cell wall. Certain Mycoplasma species are important pathogens in animal, plant and insect species. In humans mycoplasmal infections have only recently been associated with certain acute and chronic illnesses where they may function as causative agents, cofactors or opportunistic infections that cause patient morbidity. Although various Mycoplasma species are commonly found as commensals in the oral cavity and at other superficial sites, certain species can penetrate into blood and tissue and cause morbidity.
