Looking Local Novel Mutation Associated with Pain Insensitivity, Self-Mutilation, and Inability to Sweat (more detail) The NTRK1 protein (neurotrophin tyrosine kinase, receptor, type 1) is a high-affinity receptor for nerve growth factor (NGF), a neurotrophin. It has been previously established that neurotrophins and their receptors play key roles in regulating the development of both the central and peripheral nervous systems. It is now believed that the NGF/NTRK1 (neurotrophin/neurotrophin receptor) system probably plays a crucial role in the development and function of the pain/noxious stimuli reception system (the "nociceptive" system), and also in the establishment of temperature regulation via sweating in humans. In addition to being insensitive to pain, individuals with CIPA are unable to regulate their temperature by sweating. In one study of 31 affected individuals, it was reported that almost 20% of these individuals died before the age of three as the result of high fevers. CIPA was first described in 1963 in two brothers with congenital insensitivity to pain and the inability to sweat. One of these boys died after a 24-hour illness in which his temperature reached 109 °F. The autopsy of this boy revealed the almost complete absence of the first-order afferent systems considered responsible for pain and temperature sensitivity.
