Editorial Review

Product Description
In this exciting symposium, the editor brings to print important new information on AIDS and how HIV affects the brain. Each chapter focuses on one or more of the cell types that reside in or traffic through the central nervous system (CNS). Each of these cells is important to considerations of the pathogenesis of the CNS. Neurologists, AIDS physicians, and other professionals caring for AIDS patients will find that this “cell-based” view provides a unique perspective and that it will guide and stimulate future investigation of this clinically important and pathogenetically intriguing disorder. The editor also introduces some general considerations for therapeutic intervention of AIDS dementia complex (ADC).

The contributors to The Cellular Basis of Central Nervous System HIV-1 Infection and the AIDS Dementia Complex deal with the cells and mechanisms involved in HIV-1 brain infection and the resultant ADC. Each author was asked to review the involvement of their assigned cells in CNS HIV-1 infection and how these cells might be involved in the pathology and process of brain injury associated with ADC. Readers will be enlightened on the functional roles of various cells and how these cells and mechanisms might fit into the broader picture of ADC pathogenesis. ... Read more

Product Description
This volume presents in-depth reviews of the pathogenesis and clinical aspects of the AIDS dementia complex (ADC), a neurological disorder directly caused by HIV-1. The opening section examines the principal forces involved in the viral pathogenesis of ADC, i.e., HIV-1 neurotropism, immune system interactions with viruses, and cytokine circuits in the brain. An extensive section on the neuropathology and pathogenesis of ADC includes studies on the role of macrophages and microglia, the cellular neuropathology of HIV encephalitis, and the role of cytokines and amino acid excitotoxicity in mediating neuronal injury. The concluding section focuses on clinical features, evaluation, and management of HIV-related psychiatric disorders, ADC, and pediatric HIV encephalopathy. ... Read more

5-0 out of 5 stars Great Overview of the Subject
This book gives a very good over view of HIV neurology by a respected neurologist in this field.I would recommend it to any health care worker who helps people living with HIV.I like the fact it is written by one author as this makes the text more coherent than a multi-authored work which are so common these days.It is a clear and well written book. ... Product Description

Within the last decade there have been a number of new and important areas of emphasis in HIV care and research, particularly in the context of brain involvement. Without question, clinical and research innovations in the modern era of the HIV pandemic warrant an update on HIV and the brain. Written by a panel of leading experts, HIV and the Brain: New Challenges in the Modern Era offers a clear picture of brain impairment associated with the virus with particular focus on three emerging areas including different genetic strains of the virus throughout the world, interactions between advanced age and HIV, and the impact of HIV on the brain in the context of chronic, long-term treatment with highly active antiretroviral therapy (HAART). HIV and the Brain: New Challenges in the Modern Era provides a comprehensive review of both basic science and clinical implications in these important areas of the disease. The text is of particular relevance to both research scientists and clinicians involved in HIV in the modern era.

Description:
Learning and memory impairments are frequently observed in patients suffering from AIDS Dementia Complex (ADC). These effects have been linked to the presence of gp120, an HIV viral coat glycoprotein. The present study investigated the possibility that gp120 prevents the uptake of extracellular glutamate by astrocytes, leading to an interruption of the glutamate-glutamine cycle and a subsequent impairment of memory. Ten microliters of 10nM gp120 was bilaterally injected into the region of the intermediate medial mesopallium of day-old chicks at various times before, or after, training using a single-trial passive avoidance task. Gp120 was found to significantly impair memory retention when injected 10-40min after training. Memory impairments were evident within 5min of gp120 administration and remained evident 24h later. Further, the amnestic effect of gp120 could be overcome with glutamine or with precursors of glutamate synthesis, but only weakly by glutamate. These results support the conclusion that the amnestic effect of gp120 is due to an impaired uptake of glutamate by astrocytes and a subsequent interruption of glutamine supply to neurones. The data indicate that the glutamate-glutamine cycle may be a useful therapeutic target in the treatment of ADC. ... Read more
