BACE inhibitors and their use in the treatment of Alzheimer's Disease

BACE (β-site of APP cleaving enzyme) is a critical component in Alzheimer's Disease (AD), and the development of BACE inhibitors shows great potential as a therapy for the disease. BACE: Lead Target for Orchestrated Therapy of Alzheimer's Disease covers virtually all aspects of BACE from initial identification, discovery of inhibitors, and challenges in clinical development, while providing a global understanding essential for productive and successful drug discovery.

This book details the story of the discovery of BACE and its role in AD and comprehensively discusses:

  • The development of BACE inhibitors as therapeutics for Alzheimer's disease

  • The research that led to the identification of BACE

  • New BACE inhibitors currently being clinically tested

  • ADME (absorption, distribution, metabolism, excretion) and clinical trial design—topics not addressed in current field literature

  • Cutting-edge technology such as high-throughput screening, structure-based drug design, and QSAR in context of BACE inhibitors and Alzheimer's drug discovery

  • Other approaches to BACE inhibition based on interaction with the precursor protein APP

By enhancing the reader's understanding of the various aspects of the BACE drug-discovery process, this much-needed reference will serve as a key resource for all scientists involved in Alzheimer's research—and inspire new approaches to treatment of AD. ... Read more

5-0 out of 5 stars A must for anyone coping with Alzheimer's
I first became aware of this book when it was published on a smale scale for people that the authors and their colleagues encountered in their clinical work at an Alzheimer's center in New York City. I urged the authors to share this important book with a wider audeince and I am most pleased that they have done so with the publication of this paperback.

Health care for persons with Alzheimer's disease is a two-edged sword. If there is a coexisting medical condition, there is potential for great good or great harm at the hands of medical practitioners. The authors offer loads of advice about how to ensure that good is done and harm is avoided. This is a guide book that should be read once and then kept close at hand, in case of an acute illness or some other unforseen condition that requires medical attention. Mary Mitteman and Cynthia Epstein are to be commended for helping individuals and families navigate their way through the increasing complex health care maze. ... Read more

5-0 out of 5 stars Understanding Alzheimer's
John Medina, Ph.D. not only authored this detailed, concise book about Alzheimer's Disease, but also drew the terrific graphics describing the various aspects of the disease.

Medina spends quite a bit of timeexplaining the "biology of Alzheimer's Disease" which can get quitetechnical. But, if you are interested in how the brain and nerve cells areaffected, as well as other physical aspects of the disease, you will lovethese sections. His graphics are superb. My other favorite chapters dealwith understanding difficult behavior, suggesting helpful, personalinterventions, and offering practical advice for helping with the activities of daily living for a person with Alzheimer's Disease.

A glass-half-full look at Alzheimer's that reveals how to connect with someone through the fog of the disease

As many as five million Americans are living with Alzheimer's disease, and people often live with it for more than a decade. During that time, caregivers can have a vibrant and meaningful relationship with the person who suffers from the disease.

I'm Still Here focuses on connecting with someone with Alzheimer's through their abilities that don't diminish with time, such as understanding music, art, facial expressions, and touch. John Zeisel, an innovator in non-pharmacologic approaches to treating Alzheimer's, shows that people with the disease are highly creative and emotionally intelligent. Product Description

When her once-glamorous and witty novelist-mother got Alzheimer's, Eleanor Cooney moved her from her beloved Connecticut home to California in order to care for her. In tense, searing prose, punctuated with the blackest of humor, Cooney documents the slow erosion of her mother's mind, the powerful bond the two shared, and her own descent into drink and despair.

But the coping mechanism that finally serves this eloquent writer best is writing, the ability to bring to vivid life the memories her mother is losing. As her mother gropes in the gathering darkness for a grip on the world she once loved, succeeding only in conjuring sad fantasies of places and times with her late husband, Cooney revisits their true past. Death in Slow Motion becomes the mesmerizing story of Eleanor's actual childhood, straight out of the pages of John Cheever; the daring and vibrant mother she remembers; and a time that no longer exists for either of them.

5-0 out of 5 stars A wonderful book
This book came into my life at the right time. A wonderful evocative book that reflected my own experiences with family dementia. I loved the book and could not put it down, not only for the realism but also for the elegant writing. The authors love and concern for her mother showed. The effect on close relationships also well covered.

5-0 out of 5 stars Writing in Hi Def
I thought Eleanor Cooney might have something to teach me about Alzheimer's, and our loved ones who suffer with it, and caregivers who are tortured by it.Instead, she taught me about writing, and how the most gruesome and merciless life experiences can be transformed into art by a fearless writer. This book manages, against staggering obstacles, to turn the tedious misery of dementia into a vibrant emotional kaleidoscope through which the reader can experience all the shades of humor, confusion, grief, and guilt that are the constant companions of Alzheimer's.

Mary Durant, the author's mother, was nothing like my mother. Her life, full of rich experiences and vivid, fleshed-out relationships, stand as the complete antithesis of my mom's small, miserly, and tormented little world.I thought, at first, this might be an insurmountable issue for me when I began the book - the inability to RELATE.By the end of the first page, Eleanor could have told me her mother was a circus freak with webbed feet and fins and I would not have cared because I fell in love with the writing. It's that captivating.

I am about as interested in the Tang Dynasty and China as I am in how my carburetor works but I do believe I will have to read all her other books.I am now officially hooked.

5-0 out of 5 stars Powerful, Incredibly Well-Written, Deeply Touching & Helpful
Because dementia runs in my family, I intend to leave references and information for my sons.Thank you, Eleanor for sharing your incredible story with the world.

I have read the non-fiction books teaching about the black-and-white information on dementia/Alzheimer's. While researching for more "help" on Amazon, Cooney's memoir caught my eye.I gratefully read the excerpts offered online, quickly ordered it and quickly devoured it.

This is a true and gripping story -- nothing sugar-coated, which is exactly what I wanted and needed. In living REAL LIFE color, Cooney weaves her mother's past and her own throughout the memoir. I was thankful because... their lives are so much more than this one life chapter of Alzheimer's - plus, I was given some recovery time!

Eleanor Cooney shares with her reader, the unflattering but honest thoughts and feelings of a daughter with so much love, hopeful and positive intentions FOR her beloved mother.Innocently, she steps into a world for which she is unprepared and believing that love conquers all.Maybe it does for some, maybe it does eventually--this is real life and each of us will experience our own versions.

Reading this book, I felt seen, known and not alone.I also was given valuable and helpful guidance and information.

My heart and my kindness go out to Cooney and her loved ones. I thank her for bravely sharing her story. This disease will affect many of us in some way and a book such as this, offers us important insights and education. It also stretches our compassion for all who are on this path.I highly recommend it.

5-0 out of 5 stars The Hip, Cool, Funny, Brilliant, Sane Mother
Eleanor Cooney's mother, Mary Durant, and my father, Joe Thorndike, worked together at American Heritage magazine in the seventies. No one could have imagined at the time that two such lively and intelligent people could lose their minds to Alzheimer's.

Cooney makes it clear from the start that she's not going to pull any punches. She takes her first Valium eight lines into the book, and we soon discover why: her mother is difficult, and taking care of her often a nightmare. Cooney's own life is constantly put on hold as she and her partner look after a forgetful and demanding old woman. They try to do the right thing, and often they do, according to their lights. (With Alzheimer's, who can ever say exactly what the right thing is?) But sacrifice is not an idle word. The author's mother, Mary Durant, though much loved, has irrevocably disrupted their lives.

"My mother was always my favorite person," Cooney writes. "And a lot of other people's, too. Hip, cool, brilliant, funny, sane."

No wonder the author wants to protect and look after her. But what a chore it is. There are Alzheimer's caregivers--and writers--who work hard to stay cheerful. Cooney works hard to stay honest, and there are plenty of times when the job of looking after her mother overwhelms her.

"No way," she writes, "am I going to diaper my mother. We all have our limits, and this, I'm afraid, is mine."

She might avoid diapering, but she can't avoid her mother's tantrums, her demands, her extreme confusion. This is a beautifully-written book, cogent and powerful, an explicit report from the front lines of of dementia care.

5-0 out of 5 stars The shifting sands of Alzheimer's
This is a tremendously important book about one family's experience with the surreal and shifting landscape of Alzheimer's. Eleanor Cooney's story will break your heart, but also thoroughly celebrates the life of her exceptional mother, Mary Durant: a mother, writer, lover, and adventurer who manages to retain a spark of the dynamo she was up until the very last chapter.

Cooney's memoir takes a hard look at what it means to have Alzheimer's in "real world" America--with limited money, family caregivers, and no media attention or public support. Not surprisingly, there is much room for improvement. Addressing caregiver fatigue and providing affordable, quality long-term care placement for those with Alzheimer's needs to be be near the top of the list of our country's health care priorities. I would recommend this book for all medical and assistive personnel working with the elderly. ... Read more

Product Description

Alzheimer disease afflicts more than twelve million people worldwide, and its incidence is increasing at a staggering rate. People with the disorder are living longer than have those in previous generations, and they require interventions for quality-of-life issues associated with palliative care. However, the symptoms of Alzheimer disease often fail to place such persons into settings where palliative care resources are available to them. Indeed, clinicians and other caregivers may be unsure about what constitutes effective palliation in these cases. At the same time, the ethical issues involved in providing end-of-life care to persons with Alzheimer disease remain on the margins of mainstream bioethics.

In Ethical Foundations of Palliative Care for Alzheimer Disease, leading ethicists and clinicians from the United States and Europe explore ethical and scientific concerns about the diagnosis and prognosis of Alzheimer disease, challenges arising from applying palliative procedures to its symptoms, key philosophical and theological concepts central to our understanding of the disease and to end-of-life decisions, and the changing patterns of relevant medical, social, and economic policies. Cross-cultural, multidisciplinary, and state-of-the-art, this volume is a unique and important resource for bioethicists, clinicians, and policy makers everywhere.

5-0 out of 5 stars Very very helpful book!
This book had lots of good ideas for people suffering from Alzheimer's Disease. It dovetailed perfectly with another book I was given, "Creating Moments of Joy." Both books helped me create special little moments for my dad who has now passed away.

5-0 out of 5 stars A Wealth of Ideas
"What kind of activities are there for the Alzheimer's patient?" is a question that comes up often. I always recommend this book. Carmel describes all kind of activities: music, exercise, food preparation, crafts, gardening, solo activities, family games, and reminiscence. I especially like the chapter on reminiscence--with the life collage, memory book, memory box, and more. Activities are very important, as is explained in this book's introdution: "The more involved patients remain with the world around them, the more resourceful they become at finding ways to keep that world for slipping away."

Thanks tothis book, I have found dozens of creative ways to keep my mother (who is in the middle stages of Alzheimer's) involved and free from boredom. 5-0 out of 5 stars Best of the lot
Although this book is 12 years old, the content is timeless. When my mother was diagnosed with early stages of this disease, I was at a loss. I ordered two or three books on the subject, with this being one of them. This was the BEST OF THE LOT.
Easy to read and written for laymen there is not a lot of medical jargon here. Rather in concise terms signs and symptoms are listed in chapters that allow you to read about the progression of this disease. The emphasis is on the caregiver and how to relate to your loved one. This book came in so handy over the course of my caregiving, I read it over and over. It is wonderful that a nurse wrote this in such a one on one format. Although my mother is gone, this book is a keeper for dealing with other dementia patients/people one comes in contact with. I would make it mandatory reading for anyone dealing with dementia parents/patients.

5-0 out of 5 stars A wonderful resource.
My mother was diagnosed with Alzheimer's disease very early in 2000.As my grandmother had this disease also several years ago, and I cared for her also, I wanted up to date information on the disease and our options.I was the only caregiver for both my grandmother and my mother, and this book was invaluable in validating my feelings of sorrow, frustration & lonliness. It explained the disease and it's progression, also that it is NOT a bad thing if you ask for help from professionals and friends. When a person has Alzheimer's disease, the primary caretaker is usually a family member, and quite often is isolated from most outside activities. Many times, other family members and friends avoid the caretaker & patient. For myself, this disease is a slow grieving process, as you see that person leaving slowly, while still being here. This book is a great resource to help the caregiver in their life with the Alzheimer's patient.

The first time I really started to think about what Alzheimer's disease was when I read a story about Ronald Reagan. I then started to think what exactly goes into the care of someone like that? How does one provide for the needs of those afflicted with the malady?

Eileen Driscoll has writtena first rate accounting of what each person needs to know about thedealings with and caring for the Alzheimer's Patient. Driscoll's book ismore than the simple do this do that approach, it is a heartwarming storyof someone devoted to helping those much less fortunate thanherself.

What Driscoll brings to this book is a true sense of realism.Driscoll is a role model and hero and deserves to be recognized for herwork. The book gives a helping hand, and I would have to stay, from a realexpert in the field. ... Read more

5-0 out of 5 stars Caregiver

I am the caregiver for my husband who has had Alzheimer's Disease for approximately 10 years. I have made it a point to learn as much about this disease as I can. Dr. Dash's comments mirror my feelings, "...Acquiring education regarding the Alzheimer's disease process, learning strategies to deal with problematic behaviors, utilizing the resources in your community, and even successful medical management of the AD patient can help you become a more effective and healthy caregiver."This book will indeed accomplish the goal of "acquiring education." It truly covers the "nuts and bolts" of the disease. Not only is it an excellentbook for those who have been in this battle for a long time, but it will even more serve a purpose for those just starting this "long goodbye." It is an excellent addition for the home library and a good reference source. It will also enable you to be a more effective "partner" with the doctor in caring for your loved one. ... Read more

5-0 out of 5 stars Full of very practical suggestions
This book is full of practical suggestions, for example, leaving the light on in the bathroom and nowhere else at night, to suggest to the person with Alzheimer's where he/she might want to go.This has worked well for us.Very helpful for anyone dealing with Alzheimer's, and wanting to plan for the future.

5-0 out of 5 stars Making Life Easier
What an impressive book - you get 400+ pages of ways to make life more comfortable for someone with Alzheimers. These also help the caregiver in a variety of situations.
The advice covers things like how to deal with wandering and gives four types of this behavior. It tells how to eliminate places to hide and rummage, coping with fears (of bathing, of strangers, of abandonment), and managing incontinence.
The book helps with mobility issues, addresses safety and how to manage daily living. It includes a valuable section (38 pages) lists where to find special products from stair lifts to bathing aids to ramps to bedwetting alarms.
I'm so glad to see a book this comprehensive to assist with Alzheimer's needs.

5-0 out of 5 stars A caregiver's bible
Since first reading this book over 14 months ago, I've found it a constant source for information and resources. It addresses the problems I encounter on a daily basis AND gives sources for products that I can use to provide better care for my loved one.

5-0 out of 5 stars Beverly Bigtree Murphy, author, "He Used to be Somebody,"
This book is a must for caregivers and professionals facing the challenges of Alzheimer's.It is the first book I've read devoted to Alzheimer's care that offers something more than reasons to institutionalize our people.Caregivers need information, options and where to access tools andresources before they can make informed decisions about their choices ortheir role as caregiver. As someone who spent 25 years designingrehabilitations plans for handicapped people and another 10 years facingthe care of my husband, Tom, who died of this disease in our home, this isthe first volume I've picked up that speaks of caregiving in positive,life-giving, do-able terms and not the drudgery that we are all led tobelieve consumes the process. While it offers pragmatic, workable, affordable solutions to caregivers, it never loses sight of the continuedhumanity and needs of our family members for love and regard in spite oftheir loss of cognitive skills. My experience as a caregiver was met withnothing but obsticles in my quest to keep my husband home, a journey Irelate in my book, "He Used to be Somebody."I found Tom's careproblems were 75% attitude and 25% tools, resources and help. This bookoffers both to families.I highly recommend it.

5-0 out of 5 stars Much more than a how to book...
Mark Warner's book on Alzheimer's proofing your home is more than a how to book.Mr. Warner gives an in-depth description of the ways in which Alzheimer's disease affects a person's life, and the life of the extendedfamily. By understanding the disease in depth and the changes that takeplace in a person physical and emotional need, there is a context tounderstanding why modifications and alterations are needed. He explainswhat to do and why to do it.His approach is that of a loving caregiverwho not only wants to make the home safe and secure, but also wants toraise the quality of life. . Modifications are practical, but also designedto bring comfort. Even after over two decades of workingprofessionally with people with memory loss, many of his suggestions caughtme by surprise. I kept saying to myself, "why didn't I think ofthat?" He addresses all the behaviors associated with the disease, from wandering to agitation, and suggests environment solutions that couldease the situation. Not only will this bring a higher level of comfort tothe person with the disease, but many of the suggestions are a benefit foranyone who wishes to have a safe and barrier free home.I recommend thisbook to anyone caring for someone with Alzheimer's, anyone involved in homecare, and anyone who is interested in altering environments to meetspecific needs of unique individuals.Like any thoughtful book, his ideasabout adapting environments to the needs of people with Alzheimer's diseasealso stretches one's thinking about changing environments for anyone withspecific needs, or who wants to think about the highest level of safety forfamily members. ...
