The F2 populations were scored for morphology, flowering time, and rust reaction after artificial inoculation. The country's government recently announced A$77 million funding for four Cooperative Research Centers (CRCs), including the new CRC for Australian Weed Management (CRCAWM), previously known as the CRC for Weed Management Systems. The monetary support is to commence in July this year and continue through 2008. The government's announcement of the awards noted that "Weeds are amongst the most serious threats facing Australia's primary production and biodiversity with costs estimated at more than A$1.2 billion per year for grain cropping and probably at least twice that for the rest of agriculture." Extractions: We have applied PCR/DGGE to Campylobacter jejuni subtyping using a flaA PCR fragment (702bp) as polymorphic target. Thirteen different subtypes were identified in an unrelated collection of 80 Campylobacter jejuni isolates. The disciminitory Index (0.896) was slightly better than serotyping (0.868), but significant lower than PFGE and RAPD (0.974 and 0.984, respective). Improvement might be obtained by heteroduplex formation. The PCR/DGGE method does not need any restriction enzyme digestion, and the band pattern is very simple to evaluates, as the position of only a single band in the gel indicate the genosubtype. Furthermore the modified version TTGE can be automated and performed in capillarY electrophoresis. Simulation software WinMelt (BioRad) or Melt94, MUTRAV, and SQHTX (Lerman and Silverstein) are important tools to mimic the Tm melt-curves and mobility curves in denaturing gels of known DNA sequences.
