Chloroacetic acids (CAA), especially trichloroacetic acid (TCA), were considered phytotoxic secondary air pollutants of anthropogenic origin affecting conifers and were therefore intensively studied. However, another source of CAA has been then found: a forest soil in which humic substances are degraded by microbial processes to CAA and chloroform. This new aspect-formation of CAA in the soil-means that CAA are continuously produced and also biodegraded by other microorganisms in the soil and contribute thereby to the degradation of soil organic matter (SOM). Here we show for the first time the formation of dichloroacetic acid (DCA) and TCA in the forest soil by chlorination of SOM using ^3^6Cl labeled chloride. Base excision repair (BER) is a defense system that protects cells from deleterious effects secondary to modified or missing DNA bases. BER is known to involve apurinic/apyrimidinic endonuclease (APE) and DNA polymerase sz (sz-pol) among other enzymes, and recent studies have suggested that poly(ADP-ribose) polymerase-1 (PARP-1) also plays a role by virtue of its binding to BER intermediates. The main role of APE is cleavage of the DNA backbone at abasic sites, and the enzyme also can catalyze 3'- to 5'-exonuclease activity at the cleaved abasic site. Photocross-linking studies with mouse embryonic fibroblast (MEF) cell extracts described here indicated that APE and PARP-1 interact with the same APE-cleaved abasic site BER intermediate. The model BER intermediate used includes a synthetic abasic site sugar, i.e. tetrahydrofuran (THF), in place of the natural deoxyribose. APE cross-linked efficiently with this intermediate, but not with a molecule lacking the 5'-THF phosphate group, and the same property was demonstrated for PARP-1. The addition of purified APE to the MEF extract reduced the amount of PARP-1 cross-linked to the BER intermediate, suggesting that APE can compete with PARP-1. APE and PARP-1 were antagonists of each other in in vitro BER related reactions on this model BER intermediate. These results suggest that PARP-1 and APE can interact with the same BER intermediate and that competition between these two proteins may influence their respective BER related functions. ... The occurrence and distribution of the major surfactants - LAS, AES, APEOs and AEOs - and their degradation intermediates - SPCs, AP and APECs - in a marine-estuarine environment at Spain are presented. Results show that their concentration in surface sediments is clearly correlated with their usage and the existence of wastewater discharges. The degradation processes appear to lead to the formation of SPCs in the case of LAS, and to the shortening of the average ethoxylated chain length in the case of NPEOs, AEOs and AES. Vertical profiles for AEOs and AES are reported for the first time and present the highest values nearest the surface, followed by a sharp decrease with depth for all surfactants, as well as the appearance of degradation intermediates in deeper sedimentary layers. A reaction mixture containing DDT (1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane), methylene green (as the photosensitizer), and triethylamine (as the electron donor) in acetonitrile:water (1:1) was irradiated by an ordinary 150-W GE-Miser spotlight to facilitate visible-light photosensitized DDT dehalogenation. The intermediates and reaction products were identified by GC/MS (gas chromatography/mass spectrometer) and NMR (nuclear magnetic resonance). The photosensitized dehalogenation method partially degraded DDT via an electron transfer relay mechanism. Results indicate that DDT lost about three chlorines after a total of 19 days of irradiation. Aliphatic chlorines were found to be removed easier than aromatic chlorines. Various types of reductants were compared for electron donation efficiency, and photosensitizer concentration was optimized for our reaction system. In most cases, clean and simple dechlorinated products were observed. The proposed photosensitized reduction was coexisted with a thermal elimination effect for the first chlorine removal of DDT, and the photosensitized electron transfer reduction was shown to be the dominating mechanism responsible for further dechlorination after the initial stage. A sequential dechlorination pathway was proposed, with each successive dehalogenation, the reaction proceeds more slowly. The results have shown the feasibility of utilizing visible light, nontoxic dyes and electron donors to render a toxic compound less toxic and to enhance the natural carbon regeneration rates. ...
