by Ellen Glazer Carla and Marc Duffy have been reading the recent flurry of articles about cryopreserved embryos with curiosity and no small measure of frustration. The Duffy’s, busy parents of daughters 3 1/2 and 1 1/2, decided to donate their eight frozen embryos shortly after their second daughter’s birth. For them, the decision was the easy part. The hard part has been finding a medical center that wants them. Carla describes her frustration. “It feels like a don’t ask, don’t tell” kind of thing to us. Supposedly the medical centers want embryos for research, but we have yet to figure out how to donate them.” Read more of Ms. Glazer An Unlikely Crusader Becoming Our Mothers Bullish on Memories Initially, the Duffy’s assumed that there was a simple, straightforward way to give their embryos to science. Long before the potentials of stem cell research hit the news, Carla and Marc felt that “the small life of our embryos should stand for something.” That “something,” it seemed to them, was the opportunity to help others, preferably infertile couples. “We worked so hard to create the embryos,” Marko says, “We couldn’t think of not using them towards some good.”

The finding advances the effort to identify the genes involved in human male infertility, the researchers say, and suggests a possible target for a male contraceptive. From the UCSF Ancient Fly Gene Offers Insights into Male Fertility Using a human gene, UCSF scientists were able to correct a defect in infertile flies that prevented them from carrying out a key step in the creation of sperm. The finding advances the effort to identify the genes involved in human male infertility, the researchers say, and suggests a possible target for a male contraceptive. In their study of the fly, published in the January 15 issue of Human Molecular Genetics, the researchers focused on a gene that in male flies regulates meiosis, a key step in the creation of sperm and egg in all animals, including humans. In the male fly, loss of the gene, known as boule, leads to meiotic arrest, and hence infertility. Predictably, when the scientists inserted a normal copy of the gene into the flies, meiosis resumed. More notably, however, when they inserted the human form of boule, known as BOULE, into the meiosis-defective flies, development of the fly sperm also resumed.

Gene Linked To Infertility In Mice; Findings May Apply To Humans
DURHAM, N.C. A cell biologist at Duke University Medical Center has published a new study in mice that offers another possible genetic explanation for infertility in men: a gene called miwi. The research has not yet been conducted using human tissue, but Haifan Lin, an associate professor of cell biology at Duke and senior author of the study, suspects that the human counterpart of miwi, called hiwi, is probably also associated with infertility in men. The study could eventually lead to genetic testing for the human form of the miwi gene, Lin said. Piwi and miwi stem from the piwi family of genes known to be associated with the division of reproductive stem cells, which are the immature cells that can proliferate and mature to become adult sperm cells.

I wish I had words of wisdom to help you along the way...but I don't. I had only my own story to tell and the hope in my heart that YOUR quest will be more successful than mine was. *warm smiles* First is a table of links to many fertility clinics that might be helpful to you. I have not visited them, so I am not advocating or rejecting any of them. I would recommend that you research them with the American Medical Association or some other licensing agency before entrusting them with your health or hopes....or before investing your money. PS What needs to be done to understand the risks ART might have?
KH: There is an enormous and really tragic paucity of data about the outcomes of ART. There needs to be follow up done to see what effects these procedures have, and who's going to fund this kind of work is a big question. This work is expensive, and for the feds it's scary and not high on their list of things to support, because it gets dangerously close to research on embryos.
PS Yet there's such a huge debate over cloning, which raises similar issues.
KH: Right. There's an irony here that I just keep finding myself stumped over. Over the last several years, any number of groups have spent a lot of time and attention trying to figure out where they are on cloning, then advocating that position. But even in your wildest dreams, how widespread will reproductive cloning ever be? Is it going to be a real reproductive choice for my nine year-old when he decides that he's ready to build a family? Meanwhile, back at the ranch, we've got ARTs that are here and now. While any number of erudite bodies have sat around meeting tables talking about the necessity for broad public discussion about ART, there really has not been any. So that's one of our primary objectives.

Egg donation is a treatment for female infertility, which can be used to achieve pregnancy in women who lack ovaries, women with diminished ovarian function, or women who have either a genetic disease or a family history of genetic disease. The donation process begins with the retrieval of eggs from a woman with normally functioning ovaries. These donated eggs are then fertilized in a laboratory setting (in vitro fertilization) with sperm from the egg recipient’s partner or donor sperm. The resulting embryos are then transferred to the uterus of the recipient. When pregnancy is established, the egg recipient becomes the "gestational" mother and experiences pregnancy and birth. Pregnancies resulting from donated eggs to infertile women were first reported in England and from Monash University in Australia in 1983, following the development of in vitro fertilization technologies. The deficiencies of these methods led to a study of the activity of the cervix of the uterus during the cycle and to the discovery that virtually every fertile women observes, or can be trained to observe, the secretion of a particular pattern of mucus coming from the cervix around the time of fertility; this appears at the vulva as a vaginal discharge. The temperature-rhythm combination continued to be used while careful observations were made to determine those days in the cycle when it is possible for the woman to become pregnant, when she is unable to do so, and the day on which she was most likely to become pregnant. It was only after the self-observation of the mucus pattern and the application of guidelines appropriate to the desire of the couple to achieve or to avoid pregnancy in the cycle that the temperature-rhythm calculations were abandoned. By 1962 a decision was made to publish a book regarding these studies and the conclusions that had been reached. The precise time of ovulation was now able to be determined by daily measurements of oestrogen and progesterone metabolites. It was clear that ovulation occurs on the day of the Peak symptom or the following day, rarely on the second day after the Peak so that allowing for the possibility of ovum survival for 24 hours, a count of 3 days after the Peak symptom had to be applied to be sure that by the beginning of the fourth day after the Peak every woman had ovulated and the egg had disintegrated. So the earlier allowance made for the avoidance of intercourse on 3 days following the end of the mucus pattern was now more precisely translated to a count of 3 days to allow for the disintegration of the ovum following the Peak of the mucus symptom.
